Cell-Matrix Interactions
Cell-ECM interactions, through integrins expressed on the surface of hepatocytes, have been shown to modulate liver-specific function in vitro. In order to explore the effect of various ECM combinations and concentrations on hepatocellular function, we have developed a novel ECM microarray using robotic spotting . This microarray enabled us to study five matrix proteins found in the liver (collagen I, III, IV, fibronectin, and laminin) in all possible combinations with 8 replicates each (a). After seeding hepatocytes (b) and culturing for 5 days, we observed quantitative differences in intracellular expression of albumin, a marker of differentiated hepatocyte function (c-d). These results reveal a synergistic effect of ECM combinations on hepatocellular function. We are investigating the cooperation between these combinatorial effects and other factors.
